Abstract


The present investigation concerns the development and evaluation of floating tablets of Rosiglitazone Maleate which, after oral administration, are designed to prolong the gastric residence time and sustain the drug release. A 32full factorial design was employed to evaluate contribution of HPMC K15M and Xanthan gum ratio (polymer blend) and sodium bicarbonate as two independent variables and floating lag time and drug release at 12 hrs as dependent variables. The tablets were prepared by direct compression method and further evaluated for pre compression parameters, physical characteristics, in-vitro release study, Formulations were evaluated for in-vitro buoyancy and drug release study using USP dissolution testing apparatus II using pH 1.2 as a dissolution medium. The increase in drug release is observed with increase in sodium bicarbonate. Multiple regression analysis was performed for factorial design batches to evaluate the response. The release data were subjected to different models in order to evaluate their   release kinetics and mechanisms.The drug release kinetics was observed by Non- fickian diffusion mechanism. The developed floating tablets of RZM isused to prolong drug release for at least 12h.
